Toward a Functional Cure for HIV-1 Infection: The Block and Lock Therapeutic Approach
نویسندگان
چکیده
The persistence of latent, replication-competent HIV-1 proviruses in resting CD4+ T cells, and other cellular reservoirs, represents a major barrier to cure. This reservoir is impervious the immune system antiretroviral therapy, but has potential produce infectious rebound virus if therapy interrupted. There are multiple ongoing efforts identify and/or develop novel therapeutic strategies eliminate or silence this latent infection. One these termed “block lock”. “block” refers agent’s capacity inhibit (or “block”) transcription proviruses, while “lock” its induce permanent silencing typically via repressive epigenetic modifications. lock” approach elicits functional, rather than sterilizing, cure for review article focuses on approaches (i.e., small molecules, nucleic acids recombinant proteins) that have been identified block and, some cases, lock state. We also touch critical research needs be accomplished advance into humans.

Towards a Functional Cure for HIV Infection: The Potential Contribution of Therapeutic Vaccination
Human immunodeficiency virus (HIV-1) currently infects 33.3 million people globally. In 2009, 1.8 million people died from acquired immunodeficiency syndrome (AIDS) marking a decline in AIDS deaths by 19% since 1999, the estimated peak of the pandemic. This is largely due to the introduction of combination antiretroviral therapy (ART) in 1996 and its expanding access in recent years. However, d...

in fact, this study focused on the following questions: 1. is there any difference between the effect of functional/notional approach and the structural approaches to language teaching on the proficiency test of efl learners? 2. can a rather innovative language test referred to as "functional test" ge devised so so to measure the proficiency test of efl learners, and thus be as much reliable an...
15 صفحه اولToward the Development of a Single-Round Infection Assay Based on EGFP Reporting for Anti-HIV-1 Drug Discovery
Background: The rapid increase of HIV-1 strains resistant to current antiretroviral drugs is a challenge for successful AIDS therapy. This necessitates the development of novel drugs, and to this end, availability of screening systems for in vitro drug discovery is a priority. Herein, we report the modification of a previously developed system for increased sensitivity, ease of use, and cost-ef...
متن کاملRNA Interference Therapies for an HIV-1 Functional Cure
HIV-1 drug therapies can prevent disease progression but cannot eliminate HIV-1 viruses from an infected individual. While there is hope that elimination of HIV-1 can be achieved, several approaches to reach a functional cure (control of HIV-1 replication in the absence of drug therapy) are also under investigation. One of these approaches is the transplant of HIV-1 resistant cells expressing a...
متن کاملInsight into HIV-2 latency may disclose strategies for a cure for HIV-1 infection
HIV-1 and HIV-2 originate from two distinct zoonotic transmissions of simian immunodeficiency viruses from primate to human. Although both share similar modes of transmission and can result in the development of AIDS with similar clinical manifestations, HIV-2 infection is generally milder and less likely to progress to AIDS. HIV is currently incurable due to the presence of HIV provirus integr...
